Possible late-game content would be to acquire multiple (or all six?) trinkets in order to access new content. requireTrinket() would check to see the active item in the player's Trinket slot. Similar to the Elemental Planes doors, in order to pass each layer, the player needs to equip the appropriate Trinket. Beyond it would be either a map to a new boss, or a new area of the game entirely.

This would be very late-game content, as getting all currently available Trinkets is a very time-consuming process, considering many of them are rare loot drops from difficult bosses, so the reward for getting all of them should be equally powerful. Perhaps another Trinket ("Radiant Rainbow") could combine effects from all of the other Trinkets (albeit not necessarily as powerful as each individual one), since such a reward would persist across future Ascensions.

a great idea. I like the idea of Radiant Rainbow. it reminds me of the rainbow shell from chrono trigger as well as the black omen/finishing out the game. not suggesting it would have any sense of finality here, but I think it could certainly be some sort of trippy experience :D

As a note, in case a player accidentally loses any trinket once inside for whatever reason, there should be an emergency escape teleport in each section to prevent them from becoming trapped.
