After all of the parts have been soldered its time to test the board again. You need to do another continuity test on the ground plane and power traces to make sure that the soldering did not connect them. After you have made sure the ground and power circuits are separated power the board using the lab bench power supply. Use some alligator cables. Attach the ground to the ground planes and the power to the battery holder. Set the voltage to 3 volts and read what the current says. The current should read 20mA.
